Aged Dog Model
InterVivo Solutions has access to over 150 aged dogs, which naturally model the human spectrum from normal aging to early/mild Alzheimer's disease based on measures of both functional (cognition) and neuropathological (biomarkers) markers that can be used to establish efficacy. The aged dog, therefore, can be used to initially assess acute pharmacodynamic response of an NCE on AD biomarkers (such as CSF Aß42 and hyperphosphorylated tau) followed with acute or longitudinal studies assessing pharmacodynamic response of an NCE on functional (behavioral and cognitive) measures. Given the dog is a primary model for toxicology testing, the use of the aged dog can add value to a program for development of NCEs targeting AD by combining tests of efficacy with early safety markers. In this respect, the aged dog model has been useful in the development of successful AD therapeutics when used in conjunction with standard models.
In addition to establishing how behavioral effects of NCEs impact cognitive measures in dogs, InterVivo can readily obtain blood and serial CSF samples from dogs that permit our clients to assess PK-PD relationships on functional measures. In addition, the collection of these and other samples can be used to establish the PK relationship to secondary PD measures, that collectively aid in the design and interpretation of clinical trials assessing both primary and secondary markers of efficacy.
